EAN中神经抗原P2蛋白有效表位的鉴定及其与空肠弯曲菌LPS抗原性比较
Identification of dominant epitope of P2 in EAN and studying of similarity between the dominant epitope and LPS of Campylobacter jejuni
【摘要】 目的 :EAN中神经抗原 (P2蛋白 )的有效表位的鉴定以及将其与空肠弯曲菌 (Campylobacterjejuni,Cj)LPS抗原性比较。方法 :采用噬菌体呈现技术对 8个EAN模型血清抗体进行肽库筛选 ,并将获得的克隆用ELISA方法进行阳性鉴定 ,以及将阳性克隆与空肠弯曲菌LPS相似性进行比较。结果 :33个阳性克隆的亲和性均显著高于阴性对照 (P <0 0 1) ,说明它们是具有特异性的克隆 ;C J LPS与血清的亲和力显著高于对照组 (P <0 0 1) ,与阳性克隆的OD值相差不显著 ,C J LPS与阳性克隆之间存在相似性。结论 :噬菌体呈现技术可筛选出与P2蛋白具有相同表位的噬菌体克隆 ;空肠弯曲菌LPS与P2蛋白之间存在表位模拟 ,C J LPS可能是启动GBS自身反应性免疫应答的重要因素。
【Abstract】 Objective:Identification of dominant epitope of P2 in EAN and studying of similarity between the dominant epitope and LPS of Campylobacter jejuni.Methods:Screening peptide library with EAN serum.Identified positive clone by ELISA and compared similarity between positive clone and LPS of Campylobacter jejuni.Results:Affinity of all indentified positive clone increased remarkbly compared to negative control(P<0.01),suggested they might be specific clones.The affinity between EAN serum and LPS increased remarkably compared to negative control(P<0 01),but similar with positive clones.Conclusion:The result indicated that specific phage clones which have similar epitope with P2 peptide can be acquired by phage display technique;Epitope mimicry may be identified between LPS of Campylobacter jejuni and auto neural antigen,LPS may be an important factor in inducing auto reactive immune response.Epitope mimicry mechanism is a possible mechanism for instigating an autoimmune response to self antigens that in turm can lead to autoimmune disease GBS.
