ICA和PJA对高转移性人肺癌细胞体外侵袭转移能力抑制的研究

【摘要】 目的 :将新型抗癌药物淫羊藿甙 (ICA)和济南假单胞菌制剂 (PJA)作用于PG细胞 ,从肿瘤转移抑制的多个方面和环节探讨PJA、ICA抗转移作用的机制。方法 :采用粘附实验、运动侵袭实验、逆转录PCR(RT PCR)、细胞免疫组化等多种方法进行了检测。结果 :PJA、ICA可降低PG细胞对胞外基质的粘附性及侵袭、运动能力 ,减少PG细胞表面粘附分子CD44V6、LN R及胞浆内CK18的表达 ,同时细胞内c myc、Tiam 1基因mRNA水平均有不同程度的降低 ,而Nm2 3 H1mRNA水平有不同程度的升高 ,且两药有明显的协同作用。结论 :PJA、ICA通过对肿瘤转移多个步骤的抑制而发挥抗转移作用

【Abstract】 Objective: In order to further elucidate the anti metastatic mechanism of PJA、ICA, PG cells were treated with PJA、ICA in vitro Methods:In this study , adhesion assay, invasion assay, migration assay, RT PCR technique and immunohistochemical straining were used Results:PJA、ICA could decrease PG cells adhesive ratio to laminin substrate and decreased the ability of invasion or migration On the other hand, PJA、ICA reduced the expression of CD44V6、LN R and CK18 of PG cells At the same time, the relative expression levels of both c myc and Tiam 1 mRNA decreased compared with that of the control On the contrary, a tendency of the enhancement in the expression level of Nm23 H1 mRNA was also observed Also found that PJA and ICA had cooperative function Conclusion:These results suggest that PJA、ICA may play anti metastatic roles at many aspects
