P53反义RNA表达载体的构建
Construction of P53 Antisense RNA Expression Vector
【摘要】 目的 针对突变型P53 基因的致癌性 ,采用反义RNA技术以阻断癌细胞内源突变型P53 的表达 ,构建真核细胞内P53 反义RNA表达载体 .方法 采用定向克隆法将人野生型P53 基因cDNA反向插入到哺乳动物表达载体PCR3.1的EcorRⅠ和XbaⅠ位点 ,构建了反义PCR3.1 P53 (AS)表达载体 .结果 反义PCR3.1 P53 (AS)表达载体构建成功 .结论 此研究结果既为肿瘤发生提供理论基础 ,也为肿瘤基因治疗奠定基础 .
【Abstract】 Objective To study inhibition effect of P 53 antisense RNA on P 53 mutant cancer cells,P 53 antisense RNA expression vector in eucaryotic cells was constructed.Methods A human P 53 cDNA was inserted into a mammalian expression vector P CR 3.1 orientationally after using EcorR ,XbaⅠenzymes digestion to make a P 53 antisense RNA expression vector P CR 3.1 P 53 (AS).Results P 53 antisense RNA expression vector P CR 3.1 P 53 (AS) was successfully constructed.Conclusion It may provide theoretical basis for tumor etiology and gene therapy.
