NF-κB在脂肪酸合酶抑制剂诱导人结肠癌LoVo细胞凋亡中的作用

【摘要】 目的 探讨 NF-κB在脂肪酸合酶抑制剂 ( FASI)诱导人结肠癌 Lo Vo细胞凋亡中的作用。方法  FASI—浅蓝菌素处理大肠癌 Lo Vo细胞株 ,四唑盐比色试验 ( MTT法 )、片断 DNA琼脂糖凝胶电泳、流式细胞仪及荧光染色 ( AO/EB)等方法检测细胞调亡 ,用凝胶电泳迁移率法 ( EM-SA)检测 NF- κB活性。结果  Lo Vo细胞在 FASI作用下 ,细胞增殖被阻遏 ,并呈现剂量效应关系。同时诱导细胞发生凋亡。FASI对人成纤维细胞增殖无明显影响。NF- κB在不同浓度浅蓝菌素 ( 1 0 -9~ 1 0 -5)处理 Lo Vo细胞 1 2 h后 ,可抑制 NF- κB活性 ,其抑制程度呈剂量依赖性递减。结论FASI可能通过抑制 Lo Vo细胞内源性脂肪酸合成 ,诱发细胞凋亡来阻遏 Lo Vo的增殖。NF- κB可能参与 FASI诱导人结肠癌 Lo Vo细胞凋亡中的信号转导

【Abstract】 Objective To explore the role of nuclear factor KappaB(NF-κB)in fatty acid synthase inhibitor(FASI),cerulenin,induced apoptosis in human colonic cancer cells. Methods Microculture terazolium assay(MTT),agarose gel electrophoresis,fluorescence microscope(AO/EB) and flow cytometyr(FCM)assays were performed for the effects of FASI in various concentration on proliferation and apoptosis in human colonic cancer cells in vitro; electrophoretic mobility shift assay(EMSA) was used as the determination method of NF-κB activity. Results FASI,Cerulenin,inhibit the proliferation of LoVo cells in vitro and show dose response relationship. Apoptosis cells demonstrated cell nuclear DNA and apoptotic peaks and apoptotic morphology;cerulenin(10 9 ~10 5 M)treated LoVo cells in viro after 12h,NF κB activity was inhibited and dose response relationship. Conclusion The FASI, cerulenin, could induce human colonic cancer cells to die via apoptosis and suppress the growth of them by inhibition of endogenously synthesized fatty acids in cancer cells. Cerulenin could rapidly inhibit activition and translocatin of NF-κB which could be one of signaling factor for inducing apoptosis.
