血小板生成素Ⅱ对失血性小鼠死亡率的影响

【摘要】 目的 探讨血小板生成素Ⅱ (thrombopoietin ,TPOⅡ )体内的生物学活性对失血性血小板减少小鼠死亡率的影响。方法 以Babl/c纯系小鼠为实验对象 ,实验前采尾静脉血 ,了解小鼠血小板正常值。实验中通过腹腔连续 7d分组注射提纯的TPOⅡ配体Ⅰ、人工合成的TPOⅡ配体Ⅱ及重组的人血小板生成素 (rhTPO) ,并于第 7天及第 14天采眶静脉血 0 .5ml,测定小鼠外周血小板数量 ,同时观察小鼠的死亡情况。结果 用药后第 7天 ,TPOⅡ配体Ⅰ实验组小鼠外周血小板数明显高于同期阴性对照组 (P <0 .0 5 ) ,与rhTPO阳性对照组相比 ,差异无显著性 (P >0 .0 5 ) ;第 14天 ,TPOⅡ的两个实验组的血小板数均较同期阴性对照组明显升高 (P <0 .0 1) ,与阳性对照组相比 ,差异亦无显著性 (P >0 .0 5 ) ,而且随实验时间的延长 ,TPOⅡ实验组及阳性对照组血小板数均呈升高趋势。在第 7天眶静脉采血后 ,各组小鼠均陆续出现死亡 ,但TPOⅡ实验组及阳性对照组小鼠死亡率明显低于阴性对照组(P <0 .0 5 )。结论 TPOⅡ可促进小鼠外周血小板的产生 ,在一定程度上降低了失血性血小板减少的小鼠的死亡率。

【Abstract】 Objective To study the effect of thrombopoietin Ⅱ(TPOⅡ) on the exsanguine thrombocytopenia mouse death rate.Methods After the normal peripheral platelet counts were done on the samples obtained from the tail vein of purebred Babl/c mice before experiment, the purified ligandⅠof TPOⅡ, artificial compound ligandⅡ of TPOⅡand rhTPO were injected intraperitoneally once a day for 7 days. On d 7 and d 14, platelet counts were performed on 0.5 ml samples obtained from the supra orbital vein, with the condition of the mouse death monitored daily. Results On d 7, ligandⅠof TPOⅡ group platelet counts were higher than that of the negative control group ( P <0.05), while not being significantly different from that of rhTPO group ( P > 0.05). On d 14, the platelet counts of two TPOⅡ groups increased significantly as compared with the negative control group ( P <0.01), showing no significant difference from that of rhTPO group ( P >0.05). Moreover, the platelet counts of mice in two TPOⅡ groups and the positive group had shown an increasing tendency in the days following experiment. In addition, mouse death occurred in all groups of mice following their phlebotomy from the supraorbital vein on d 7. But the death rate of negative control group was evidently higher than that of any other groups ( P <0.05).Conclusion TPOⅡ′s biological activity obviously increases platelet production, thereby reducing the exsanguine thrombopenia mouse death rate.
