肿瘤坏死因子-α对血管平滑肌细胞核转录因子κB活性的影响
Effect of tumor necrosis factor-α on NF-κB activity in cultured smooth muscle cells
【摘要】 目的 探讨肿瘤坏死因子 α(TNF α)对血管平滑肌细胞 (SMC)核转录因子 κB(NF κB)活性的影响。方法 选择大鼠主动脉SMC经用含 0 5 %小牛血清的DMEM培养 4 8h后 ,加入 2 5ng/ml的TNF α入培养基中 ,于 0、15和 3 0min收集细胞 ,提取胞核蛋白。电泳流动漂移技术 (EMSA)检测NF κB活性的变化。结果 静息状态下的SMC具有一定的NF κB活性水平。经TNF α刺激后 ,其活性迅即增加。密度扫描结果提示 ,NF κB活性 15min时为对照 2倍 ,3 0min为对照 3倍。结论 TNF α使血管SMCNF κB活性增加 ,提示TNF α对SMC的调节作用部分是通过NF κB激活通路发挥的。
【Abstract】 Objective To investigate the effect of tumor necrosis factor-α(TNF-α) on nuclear factor-κB(NF-κB ) activity in cultured vascular smooth muscle cells (SMCs). Methods Rat(n=12) aortic SMCs were cultured in DMEM containing 0.5% bovine calf serum for 48 hours and then 25ng/ml of TNF-α was added into the medium for stimulation.Cells were collected and nuclear protein of cells was extracted at 0, 15 and 30 minutes following stimulation. NF-κB activity in stimulated cells was examined by electrophoretic mobility shift assay(EMSA). Results Baseline NF-κB nuclear binding in SMCs increased more than two-fold at 15 min (n=4, P<0.01) and three-fold at 30 min(n=4, P<0.01) following stimulation of TNF-α. Conclusions TNF-α could enhance transient mobilization of NF-κB in vascular SMCs, which suggested that some regulating effect of TNF-α on vascular SMCs might be exerted through NF-κB activation pathway.
