人蛔虫变应原致喘豚鼠支气管肺泡灌洗液中嗜酸性粒细胞形态测量分析
Morphometrical analysis of the eosinophils in the bronchoalveolar lavage fluid of asthmatic guinea pigs provocated by Ascaris Lumbricoides allergen
【摘要】 目的 了解变应性哮喘过程中嗜酸性粒细胞 (EOS)的形态学变化。方法 用多功能检测系统 ,对人蛔虫全虫变应原激发后不同时间段致喘豚鼠支气管肺泡灌洗液中的EOS进行形态学测量分析。结果 激发后 1h致喘豚鼠气道中已有EOS浸润 ,激发后 2 4h组EOS数最多 ,72h组其次 ,各实验组的EOS数均明显大于正常对照组 (P <0 .0 1) ;激发哮喘后EOS胞浆量逐渐增多 ,致喘后 2 4hEOS胞浆量明显多于正常对照组 ,其体积亦大于对照组 ,而体密度和核浆比则明显小于正常对照组 ,但致喘后 72hEOS胞浆量又明显减少 ,基本恢复到正常对照组水平 ;72h组EOS胞浆面积小于 1h组和 2 4h组 ,其体积明显小于 2 4h组 ,而EOS体密度和核浆比则明显大于 1h组和 2 4h组 ,但其胞浆投影面积、体密度和核浆比均与正常对照组无明显差异 ;各实验组与对照组间EOS核体积没有明显变化。结论 迟发相哮喘反应中豚鼠气道内EOS浸润程度较速发相反应重 ;EOS脱颗粒发生在哮喘激发后 2 4h以后 ,EOS形态学发生显著变化
【Abstract】 The eosinophils(EOS) in the bronchoalveolar lavage fluid(BALF) of guinea pig model of allergic asthma were analyzed morphometrically. A normal guinea pig which was unimmunized and unprovocated was also analyzed morphometrically. The results showed that the eosinophil number, the volume of EOS and the projection area of cytoplasm increased with time from 1 hour to 24 hours after asthma provocation. However from 24 hours to 72 hours, both of them decreased significantly. On the contrary, the density of nuclear(Vv) and the ratio of nuclear to plasma(Rnp) decreased with time from 1 hour to 24 hours, while from 24 hours to 72 hours both of them increased dramatically. Seventy two hours after asthma provocation, the volume of EOS, the projection area of cytoplasm, Vv and Rnp returned to normal state. Both the projection area and volume of nuclear had no significant change all through the observation time. Moreover, numerous eosinophilic granules were observed around eosinophils in 72 hours group. These results suggested that the eosinophil infiltration in delayed type asthma was much more serious than that in immediate type asthma and the release of eosinophil granules began later than 24 hours after asthma provocation.
