乳腺癌p21 WAF1基因多态性及其临床意义
p21 WAF1 gene polymorphisms and it’s clinical implication in breast carcinoma
【摘要】 目的 探讨乳腺癌组织 p2 1WAF1基因多态性及其临床意义。 方法 采用聚合酶链反应单链构象多态性技术 (PCR SSCP)法检测 10 0例乳腺癌组织 p2 1WAF1基因的结构变异 ,同时应用免疫组织化学SP法检测乳腺癌组织p2 1WAF1的蛋白表达。结果 5 % ( 2 /4 0 )的对照组组织出现多态性 ,18% ( 18/10 0 )的乳腺癌组织出现两种多态性 ,两组间差异有显著性 ( χ2 =30 .94,P <0 .0 5 ) ;p2 1WAF1基因多态性与蛋白表达 ( r =0 .5 76,P <0 .0 1)和组织学分级 (r =0 .360 ,P <0 .0 1)呈正相关。结论 p2 1WAF1基因多态性在乳腺癌组织中明显增加 ;p2 1WAF1基因多态性可能与促进细胞增殖和增强对癌的易感性有关。
【Abstract】 Objective To investigate the p21 WAF1 g ene polymorphisms in human breast carcinoma and its clinical implication. Methods Using PCR-SSCP technique, the p21 WAF1 gene poly mo rphisms were detected in 100 cases of breast carcinomas and 40 control samples o f benign breast diseases, and the expression of p21 WAF1 protein was detected by immunohistochemistry method. Results Two p21 WAF1 gene p olymorphisms were found in 18 % (18/100) of breast carcinomas and 5 %(2/40) of c ontrol samples (χ~2=30.94,P<0.05). Both p21 WAF1 gene polymorphisms wer e cor related with expression of p21 WAF1 protein(r=0.576,P<0.01)and with hi stolo gical grade of breast carcinomas((r=0.360,)P<0.01). Concl usion The occurrence of p21 WAF1 gene polymorphisms is apparently increased i n breast carcinomas. The polymorphisms of p21 WAF1 gene might be associated with advanced cellular proliferation and increased susceptibility for carcinoma.
