中暑士兵热应激蛋白71基础和诱导水平的分析

【摘要】 目的 分析中暑与对照士兵外周血淋巴细胞热应激蛋白 71(HSP71)基础和诱导性水平的差异 ,探讨中暑发生的分子机制。方法 采用流式细胞术方法检测两组士兵外周血淋巴细胞HSP71。结果 基础状态下 ,对照和中暑士兵HSP71水平 (对数平均荧光强度分别为 2 6 8.10、390 .0 5 )均较低 ,且差异无显著性 (P >0 .0 5 ) ;在体外 37℃培养条件下 ,淋巴细胞HSP71水平较基础状态明显上升 ,中暑士兵HSP71水平 (对数平均荧光强度为 140 5 .0 8)在此条件下较对照士兵 (对数平均荧光强度为 931.16 )更高 ,差异有显著性 (P <0 .0 1) ;在 41℃应激 1h后 ,对照组士兵淋巴细胞HSP71平均水平轻度升高 (对数平均荧光强度为 95 5 .99) ,而中暑组HSP71不但没有升高 ,反而降低 (对数平均荧光强度为 1148.92 )。结论 中暑士兵外周血淋巴细胞HSP71诱导性发生改变 ,提示中暑士兵HSP71基因及其表达存在差异性 ,其热耐阈下降 ,热耐受能力不能正常形成。

【Abstract】 Objective To analyse the basal and inducible levels of heat stress protein 71(HSP71) in peripheral blood lymphocytes in heat illness and control soldiers,and to explore the mechanism of heat illness. Method Flow cytometry was used for detecting HSP71 in peripheral blood lymphocytes. Results In the basal condition,levels of HSP71 were low and no significance was found between control and heat illness soldiers[268.10 and 390.05 log of mean fluoresence intensity(logMFI) respectively].After culture at 37 ℃ for 5 h,the level of HSP71 in both heat illness and control soldiers remarkably increased compared with basal condition.The level of HSP71 of heat illness soldiers was much higher than control(1 405.08 logMFI and 931.16 logMFI, P <0.01).HSP71 in control soldiers increased slightly after stress at 41 ℃ for 1 h(955.99 logMFI),while in heat illness group,HSP71 declined instead of reaching a higher level(1 148.92 logMFI). Conclusion The inducibility of HSP71 changed in heat illness soldiers.It suggests that the individual differences of hsp71 gene and expression may play a role in the occurrence of heat illness.The threshold of thermotolerance decreased and the ability of thermotolerance could not be normally formed.
