【摘要】 目的探讨肝移植病人 TNF-α基因多态性与急性排斥反应的关系。方法 PCR 特异序列引物法(polymease chain reaction sequence-specific primers method)检测121例肝移植病人 TNF-α基因启动子308位点基因型。结果 TNF-α基因启动子-308位点,G/G 与 G/A 基因型间的急性排斥反应发生率,无明显差异(26.39％,24.49％;P>0.05);并且,两种基因型间的肝移植病人1年、1～5年及5年以上生存率均无显著差异(P>0.05)。结论 TNF-α基因多态性与肝移植病人术后急性排斥反应及生存期无确切关系。更多还原

【Abstract】 Objective To explore the relationship between TNF-α gene polymorphism and acute rejection after liver transplantation.Methods The relationship between TNF-α gene polymorphism and occurrence of acute rejection was investigated in 121 adult liver transplant recipients.All patients received standard immunosuppression with corticosteriods,azathioprine and cyclosporine or FK506. Thirty-one patients (25.62%) developed acute rejection 1-18 weeks following the transplantation. PCR sequence-specific primers were used to determine genotypes and TNF-α gene promoter-308 posi- tion polymorphism (G>A).Results The occurring rate of acute rejection in liver transplant patients with G/G and G/A genotypes at TNF-α gene promotor-308 position was 26.39% and 24.49%,re- spectively (P>0.05).Meanwhile,the survival rate in liver transplant patients with G/G and G/A genotypes at TNF-α gene promotor-308 position was 12.5% and 24.49% for 1 year,68.06% and 65.31% for 1—5 years as well as 19.44% and 10.20% for more than 5 years,respectively (P> 0.05).Conclusions After the liver transplantation,there is no relationship between TNF-α gene pol- ymorphism and acute rejection or survival rate.更多还原
