α辐射诱发人胚肺细胞转化及DNA链断裂
Relationship between cell transformation and DNA strand breaks induced by α-radiation
【摘要】 目的 探索α辐射所致DNA链断裂损伤与辐射致癌之间的定量关系。方法 用缺口翻译技术检测DNA链断裂 ,用软琼脂克隆形成率评价细胞转化。结果 0 2 5~ 5Gyα粒子照射后的人胚肺细胞 ,DNA链断裂损伤和半固体琼脂培养集落形成率均明显增加 ,并显示出良好的剂量效应关系。进一步分析表明 ,各受照射组细胞DNA链断裂与细胞转化率之间存在着显著的正相关。结论 α辐射致癌与辐射致细胞DNA链断裂损伤密切相关
【Abstract】 Objective\ To inverstigate the quantitative relationship between HEL 9402 cell transformation and DNA strand breaks induced by α particles. Methods\ Nick translation assay was used to detect the level of DNA strand break.Cloning formation efficiencies in soft agar were used to evaluate cell transformation. Results\ Level of DNA strand break and cloning formation efficiencies in soft agar increased obviously after HEL 9402 cells were irradiated by 0.25-5 Gy of α particles,and there were statistical relationships between DNA strand breaks and cell transformation of irradiated HEL 9402. Conclusion\ DNA damages are closely related to α particle irradiation induced carcinogenesis.
