【摘要】 目的　研究发现控制自然杀伤 (NK)细胞低活性的基因与HLAⅠ类基因相关联。在此基础上 ,检测肺癌病人NK细胞活性和HLAⅠ类抗原 ,以期发现肺癌的HLAⅠ类基因分布的特点及其与NK细胞活性的关系。方法　采用国际标准的微量细胞毒分型方法对 46例肺癌病人和 34例正常人对照进行HLA A、 B分型 ;采用乳酸脱氢酶释放法测定其NK细胞活性 ;数据的统计学处理采用t′检验及卡方检验或Fisher精确概率法。另外 ,46例肺癌病人与已知其单倍型的96例正常人进行了传统的疾病关联分析。结果　发现肺癌病人的NK细胞活性 (4 1.87± 2 5 .44 )明显低于对照组 (6 2 .6 5±47.5 0 ) ,t′=2 .32 ,P <0 .0 5。肺癌病人NK细胞活性下降与HLA的型别分布无关 ,肺癌与HLA A , B抗原均无显著关联。结论　肺癌的发生及其自然杀伤细胞活性下降与HLAⅠ类无关联更多还原

【Abstract】 Objective To explore the association of the distribution of HLA class Ⅰ in lung cancer paitents with the occurrence of lung cancer and the lower natural killer activity found in lung cancer patients .Methods 34 healthy individuals,aged 25～26 yrs,10 men and 24 women,and 46 lung cancer patients including 19 squamous cell carcinoma,22 adenocarcinoma,5 small round cell carcinoma,aged 36～73 yrs,38 men and 8 women were included.Their blood samples were taken before treatment of radiotherapy,chemotherapy and operation.96 random individuals were as the control group.46 patients and 34 normal individuals were typed for HAL-A,-B antigens by a microcytotoxicity assay and tested for natural killer cell activity by lactate dehydrogenase(LDH) enzyme release assay.Results The frequencies of HLA-A,-B alleles in cancer patients were not different from those of 96 normal controls.Compared with 34 normal individuals(62.65±47.50),the lung cancer patients showed a lower level of NK cell activity(41.87±25.44)(t′=2.32 P<0.05).The number of NKB homozygous in patients was no more than that in normal individuals and the distribution of the low NK activity in different HLA-B type complex was not different between patients and normal individuals.Conclusion The occurrence of lung cancer and the lower NK activity found in lung cancer patients are not associated with HLA class Ⅰ.更多还原
