健骨二仙丸对去势大鼠骨结构和血清 IGF-Ⅰ的影响

【摘要】 目的 :观察健骨二仙丸对去势大鼠骨结构和血清胰岛素样生长因子 1 ( IGF- )水平等指标的影响。方法 :1 2月龄雌性 SD大鼠随机分为空白对照组 ( OVX)、雌激素对照组 ( OVX+ E)、健骨二仙丸 1、2组( OVX+ J1、J2 ) ,卵巢切除后 7天开始给药 ,6月处死大鼠检测血清 IGF- 水平、股骨干重、灰重和骨钙含量 ,骨矿化率等。结果 :OVX+ J1、J2组血清 IGF- 水平显著高于 OVX组和 OVX+ E组。 OVX+ J1、J2组、OVX+ E组股骨干重、灰重、骨钙含量和骨矿化率显著高于 OVX组。 OVX组骨小梁明显稀少 ,同样倍数下骨小梁的连接中断点多 ,骨小梁明显变细 ,间距加宽 ,甚至在部分区域出现断裂或较大的空白区域。 OVX+ J1、J2组和 OVX+ E组骨小梁排列密集 ,互相连接成网状 ,骨小梁厚度大 ,其间距小 ,且三组间无明显差异 ;结论 :健骨二仙丸具有抑制骨吸收 ,防止骨丢失 ,维持骨组织的正常结构和力学特性的作用 ,其作用机制可能与提高血清IGF- 水平有关

【Abstract】 Objective:To determined the effects of ’JianGuErXianWan’ on bone microstructure and serum insulin like growth factor 1(IGF Ⅰ) in the ovariectomized rat. Methods:Forty female Sprague Dawley rats were randomly divided into four groups: Control group (OVX group), Conjugated estrogen group(OVX+E group), and JIANGUERXIANWAN 1,2 groups (OVX+J1, J2 groups) , and the drugs or placebo was given per oral daily for 6 months from day 7 of having been ovariectomized. and the serum IGF Ⅰ, the femur degreased dry weight, ash weight and calcium content, and the Mineralization Contents (%) of tibia were tested 6 months later after the rats were killed.Results:Serum IGF Ⅰ, femur degreased dry weight, ash weight and the content of bone calcium of femur, and the Mineralization Contents (%) of OVX+E group and OVX+J groups are significantly higher than that of OVX group. Histomorphological researches show that OVX group rats are osteoporotic as evidenced by the presence of thin bone trabecula, small individual islands of cancellous bone with loss of connectivity between trabeculae. In comparison, bone of OVX+J groups and OVX+E group animals exhibit numerous, well-connected trabeculae. Conclusion: These results indicate that ’JianGuErXianWan’ can effectively inhibit the bone resorption, prevent the bone loss, and maintain the normal structure and mechanical characteristics. The mechanism of the drugs on the osteoporosis may be related to the rising serum IGF Ⅰ level.
