西洋参叶20S-原人参二醇组皂苷对急性心肌梗死犬血流动力学和氧代谢的影响
Effects of Panax quinquefolium 20s protopanaxdiol saponins on the hemodynamics and cardial oxygen metabolism in dogs with acute myocardial infarction
【摘要】 目的 研究西洋参叶 2 0S 原人参二醇组皂苷 (Panaxquinquefolium 2 0s protopanaxdiolsaponins ,PQDS)对急性心肌梗死犬血流动力学和氧代谢的影响。方法 采用麻醉开胸犬结扎左冠状动脉前降支 (LAD)产生急性心肌梗死 (AMI)模型 ,测定AMI6h犬的心脏血流动力学及心肌氧代谢参数。结果 iv滴注PQDS 10 ,2 0mg·kg-1,均能明显增加心肌血流量 ,降低冠脉阻力 ;亦可明显减慢心率 ,降低血压 ,LVP ,+dp/dtmax、LVWI及TPR ,增加SI及CI;并明显减少 -dp/dtmax降低及LVEDP升高幅度 ;同时明显降低心肌耗氧量、心肌氧摄取率及心肌耗氧指数。结论 PQDS主要通过减少左室作功 ,降低心肌耗氧量 ,增加缺血心肌供血等环节发挥抗心肌缺血作用
【Abstract】 OBJECTIVE To study the effects of Panax quinquefolium 20 S protopanaxdiol saponins extracted from the leaves of panax quinque folium (PQDS) on the hemodynamics and cardial oxygen metabolism in dogs with acute myocardial infarction (AMI).METHODS The parameters of hemodynamics and cardial oxygen metabolism were determined by using the model of ligation of LAD in the anaesthetized open chest dogs.RESULTS In dogs treated with PQDS (in a dosage of 10 and 20 mg·kg -1 iv infusion),the myocardial blood flow (MBF) was increased and coronary vascular resistance (CVR) was decreased significantly.The heart rate (HR) was slowed.The arterial blood pressure (ABP),left ventricular pressure (LVP),the maximum rise rate of left ventricular pressure(+dp/dt max ),left ventricular work index (LVWI) and total periphery resistance(TPR) were reduced,whereas the stroke index(SI) and cardiac index (CI) were increased.In addition,the decreasing range of the maximum decline rate of left ventricular pressure(-dp/dt max ) and the rising range of left ventricular end diastolic pressure (LVEDP) were reduced.Meanwhile the cardiac oxygen consumption,myocardial oxygen utilization rate and cardiac oxygen consumption index were also decreased.CONCLUSION PQDS had protective effects on acute myocardial ischemia including improving heart function,decreasing cardiac oxygen consumption and increasing myocardial blood flow,etc.
