新型镇静催眠药扎来普隆的合成工艺改进
Improvement on the Synthetic Technology of Zaleplon as a New Sedative and Hypnotic Drug
【摘要】 以苯乙酮为原料 ,经硝化、还原、酰化、缩合、烃化、环合反应合成了新型镇静催眠药扎来普隆 ,总收率为 2 2 1%。其结构经1H NMR、IR、MS得到确证。在烃化反应中 ,采用溴乙烷代替价格昂贵的碘乙烷 ,收率达 90 % ,降低了成本 ,适于工业生产
【Abstract】 Zaleplon,a new sedative hypnotic drug was synthesized from acetophenone by nitration,reduction,acylation,condensation,hydrocarbylation,and cyclization.The overall yield was 22 1%,and the structure was confirmed by 1H NMR,IR and MS.In the hydrocarbylation reaction,bromoethane was used instead of the expensive iodoethane with the yield of 90%,and the productive cost was reduced and it is more suitable for the industrial pruduction.
