ZnMT对甲基汞致小鼠肝细胞DNA损伤的拮抗研究
Resislance of Zinc - metallothionein on DNA Damage of Mouse Livers Induced by Methylmercury
【摘要】 目的 研究锌金属硫蛋白(ZnMT)对甲基汞致小鼠肝细胞DNA损伤的影响。方法 采用单细胞凝胶电泳(single cell gel electrophoresis,SCGE)技术检测锌诱导机体产生ZnMT及体外给予纯品ZnMT对甲基汞致小鼠肝细胞DNA损伤的影响。结果 体内给予小鼠5mg/kg的锌剂及在培养基中加入终浓度为0.01μmol/L的纯品ZnMT均可以减轻甲基汞对小鼠肝细胞DNA损伤程度,且损伤程度越轻,拮抗作用越明显。结论 ZnMT可以拮抗甲基汞引起小鼠肝细胞DNA的损伤。
【Abstract】 Objective To study the effects of ZnMT on DNA damage induced by methylmercury in mouse livers Methods We used the single cell gel electrophoresis (SCGE) to explore the protection of ZnMT against DNA damage induced by methymercury in vivo and in vitro. Results ZnMT decreased tlie DNA damage induced by metliylmercury in mouse liveisIt included synthesis of ZnMT treated with 5mg/kg zinc in vivo and 0.01μmol/L ZnMT m vitro .The lower of DNA damage induced by MeHg, tlie more extremely pro-tected. Condusions ZnMT can protect the DNA damage induced by MeHg in mouse livers.
