抗牛布鲁氏菌独特型抗体杂交瘤细胞(F9)的生长和代谢

【摘要】 用RPMI16 40培养基对抗牛布鲁氏菌独特型抗体杂交瘤细胞株 (F9)进行培养 ,并对其生长代谢进行测定。该细胞不论接种 1.1× 10 5cells/ml还是 2 .2× 10 5cells/ml,其增殖最高倍数只能达 2~ 3倍 (3.2 2× 10 5cells/ml/ 48h和 3.7× 10 5cells/ml/2 4h)量 ,葡萄糖在 48~ 72小时内全部耗尽 ,共产生大量乳酸 ,随着葡萄糖的耗竭 ,碳源的消失 ,乳酸的积累 ,培养液pH急剧下降 ,致使细胞活力受到很大影响 ,细胞几乎无维持期就快速死亡。

【Abstract】 A hybridoma cell line(F9),which secreted anti_bovine brucellosis idiotypic antibody,was cultured with RPMI medium 1640 and then determined its growth and metabolism.Inoculating the cell with 1.1×10 5 cells/ml and 2.2×10 5 cells/ml,its highest output could only increase 2~3 times[3.22×10 5 cells/ml(48h)3.7×10 5 cells/ml(48h)].Glucose was depleted in 48~72 hours and meanwhile produced a mass of lactic acid[1.48g/L(72h)and 1.65g/L(48h)].With using up glucose,disappearing of carbon sources and accumulating lactic acid,the pH of medium sharply declined.These factors affected on the cell viability and resulted in the cells died immediately,hardly without maintain period.
