剑麻细胞悬浮培养诱导蛋白酶的研究

【摘要】 本研究将经过高浓度生长素预培养的剑麻愈伤组织细胞.接种入LS、MS、N6、B5四种液体培养基中,在30d培养过程中动态每2-5d测定培养细胞的生长速率、剑麻蛋白酶活力和培养液的pH值变化。结果表明:悬浮培养细胞的生长量成倍地高于固体培养细胞;LS、MS培养基是剑麻细胞悬浮培养及诱导产蛋白酶最适的培养基;悬浮培养液中的蛋白酶活力占细胞蛋白酶活力的20.0%-46.9%;悬浮培养细胞生长速率与培养液的pH值存在一定的相关性。

【Abstract】 Before suspension culture, the callus of Agave sisalana was cultured in high concentration of 2,4-D mediums for a certain time, then translated it into the suspension medium of LS、MS、N6 and B5. During this 30 days cultural time, the callus growth rate, Agavain-SH activity and pH value of suspension medium were tested every 2 to 5 days. The results showed that in suspension medium the callus growth was two times higher than that in solid medium. LS and MS mediums were the best mediums for the callus growth and inducement of Agavain-SH. There were 20.0% to 46.9% of Agavain-SH secreted to the suspension medium. There are a certain interrelation between the callus growth rate and the suspension pH value.
