【摘要】 目的 研究西洋参叶 2 0 S-原人参二醇组皂苷 ( PQDS)对急性心肌梗死大鼠交感神经递质及肾素 -血管紧张素系统 ( RAS)的影响。方法 通过大鼠结扎左冠状动脉前降支 ( L AD)急性心肌梗死模型 ,测定心肌梗死面积 ,血清肌酸磷酸激酶 ( CK)及乳酸脱氢酶 ( L DH)活性 ,血清去甲肾上腺素 ( NE)及肾上腺素 ( E)含量 ,血清血管紧张素转化酶 ( ACE)及血浆肾素 ( R)活性。结果  PQDS12 .5 ,2 5 ,5 0 m g/ kg舌下静脉给药 ,对急性心肌梗死 2 4 h大鼠可明显缩小心肌梗死面积 ,降低血清 CK及 L DH活性 ,并明显降低血清 NE、E含量及血清 ACE和血浆 R活性。结论 PQDS对急性心肌缺血具有保护作用 ,可能与其抑制交感 -肾上腺髓质过度兴奋 ,减少儿茶酚胺 ( CA)大量分泌及其抑制 RAS激活 ,减少血管紧张素 ( Ang )生成 ,打破 CA与 RAS相互促进造成的恶性循环等机制有关。

【Abstract】 Object To study the effect of Panax quinquefolius L. 20S-protopanaxdiolsaponins derived from leaves of P. quinquefolius (PQDS) on sympathetictransmitter and renin-angiotesin system (RAS) in rats with acute myocardial infarction. Methods Rat acute myocardial infarction models were prepared by ligating left anterior descending coronary artery (LAD). The changes of myocardial infarct size, serum creatine phosphokinase (CK), lactic dehydrogenase (LDH), serum noradrenaline (NE), adrenaline (E), contents, serum angiotensin converting enzyme (ACE) and plasma renin activities were determined. Results The myocardial infarct size, serum CK and LDH activities, noradrenaline and adrenaline contents were significantly reduced, after iv injection through sublingual vein 12.5, 25, and 50 mg/kg of PQDS, 24 h after myocardial infarction. The serum ACE and plasma renin activities were also decreased markedly. Conclusion PQDS showed protective effect on acute myocardial ischemia, possibly due to its action to inhibit hyperesthesia of sympathetic-adrenal medulla, by decreasing catecholamines (CA) level and angiotensin Ⅱ(Ang Ⅱ) production and interferes the vicious cycle caused by synergic actions between CA and RAS.
