HPLC测定山豆根中苦参碱的含量

【摘要】 目的 :建立山豆根苦参碱含量的测定方法。方法 :采用苯基柱 ,甲醇 水 三乙胺 (5 0∶5 0∶0 .0 5v/v)为流动相 ,等度洗脱 ;流速为 1.0ml·min-1;紫外检测波长 2 19nm ;柱温为 30°C。结果 :最低检测限为 2ng·ml-1;加样回收率在 95 %以上 ,日内及日间的相对标准偏差分别小于 2 %和 4% ;线性范围 1.0~ 10 0 0 μg·ml-1。结论 :该方法简单 ,快速 ,重复性好

【Abstract】 Objective: To establish the method for determining matrine in Sophora subprostrala Chun et T.Chen. by HPLC. Methods: A CLC-phenyl column was selected as separation column at 30 °C . Matrine was isocratically eluted by methanol-water-triethylamine (50∶50∶0.05,v/v) at flow rate of 1.0ml·min -1 . The peak of matrine was monitored at UV 219nm. Results: The detection limit was 2ng·ml -1 . The recovery of the added sample was above 95%. RSD of intra-day and inter-day was less than 2% and 4%, respectively. The linear range was 1.0~1000μg·ml -1 . Conclusion: The method was simple, rapid and producible.
