清开灵对内毒素性发热家兔下丘脑cAMP及腹中隔区AVP含量的影响

【摘要】 目的 :探讨清开灵 (QKL)注射液的解热机制。方法 :建立家兔内毒素 (ET)性发热模型 ,观察清开灵对家兔体温的影响和用放免法检测下丘脑cAMP及腹中隔区AVP含量的变化。结果 :①ET组的△T为 (1 6 8±0 46 )℃、TRI6 为 2 9 5 9± 10 39、下丘脑cAMP含量为 (2 90± 0 40 )nmol g、腹中隔区AVP含量为 (47 32± 3 77)ng g ,分别显著高于NS组的△T[(0 0 8± 0 11)℃ ]、TRI6 (- 0 .15± 4 2 9)、下丘脑cAMP含量 [(1 0 5± 0 37)nmol g],腹中隔区AVP含量 [(11 78± 7 87)ng g]及QKL +ET组的△T[(0 48± 0 18)℃ ]、TRI6 (13 71± 3 2 9)、下丘脑cAMP含量 [(1 37± 0 2 3)nmol g]、腹中隔区AVP含量 [(2 5 2 4± 2 6 1)ng g](P <0 0 1)。②四组下丘脑cAMP含量变化与体温变化呈明显正相关 (r=0 .90 4,P <0 0 1)。结论 :清开灵的解热机制可能是通过抑制下丘脑cAMP生成 ,同时通过腹中隔AVP而发挥作用。

【Abstract】 AIM: To investigate the mechanism of Qing-Kai-Ling(QKL) Injecta suppressing endotoxin(ET)-induced fever in rabbits.METHODS: ①The ET-induced fever model was established in rabbits. QKL injecta was administered intravenously, and the febrile response of rabbits was observed. The cAMP content in the hypothalamus (HP) and arginine vasopressin (AVP) content in the ventral septal area (VSA) were determined by radioimmunoassay.RESULTS: ①The maximal increment in body temperature (△T)[(1 68±0 46)℃],6 h thermal response index (TRI 6)(29 59±10 39), cAMP content in the HP[(2 90±0 40) nmol/g] and AVP content in the VSA[(47 32±3 77) ng/g] in the ET group were significantly higher than those in the normal saline (NS) group[(0 08±0 11)℃,-0.15±4 29,(1 05±0 37) nmol/g,(11 78±7 87) ng/g, respectively] and the QKL+ET group[(0 48±0 18)℃, (13.71±3.29), (1 37±0 23) nmol/g,(25 24±2 61) ng/g, respectively].②The positive correlation was observed between the cAMP content in the HP and the fluctuation of body temperature ( r=0.904,P<0 01 ).CONCLUSION: The antipyretic mechanisms of QKL were probably due to inhibiting the increase in cAMP content in HP, and meanwhile stimulating the release of AVP in VSA.
