CD68、CD43和肌动蛋白在实验性家兔动脉粥样硬化斑块中的表达
Expression of CD68、CD43 and Actin in Experimentally Atherosclerotic Plaques in Rabbits
【摘要】 目的 :探讨动脉粥样硬化斑块中泡沫细胞的来源及其与斑块形成的关系。方法 :应用免疫组织化学S -P法对实验性家兔主动脉和冠状动脉动脉粥样硬化斑块 (脂纹期 6 2个和纤维斑块期 38个 )中CD6 8、CD43和Actin的表达进行检测。结果 :在主动脉和冠状动脉斑块中 ,CD6 8、CD43和Actin的表达无明显差异 (P >0 .0 5 )。脂纹期斑块中 ,CD6 8阳性物质弥散分布 ,而CD43阳性物质主要分布在斑块的中央 ;两者的表达显著大于纤维斑块期 (P <0 .0 1)。纤维期斑块中Actin阳性物质主要分布在斑块的基底部和纤维帽中 ,其表达显著高于脂纹期 (P <0 .0 1)。结论 :动脉粥样硬化脂纹期斑块的泡沫细胞主要是单核巨噬细胞源性 ;纤维斑块期主要是血管平滑肌细胞源性 ,其与形成纤维帽有关。局部浸润的T淋巴细胞与巨噬细胞相互作用可能是斑块中脂质坏死的重要因素
【Abstract】 Objective To investigate the relationship between the origins of foam cell and the formation of the atherosclerotic plaques.Methods The expression of CD68,CD43 and actin were examined by immunohistochemical S-P method with monoclonal antibodies in aortic and coronary plaques of experimental atherosclerosis (including 62 fatty streak plaques and 38 fibrous plaques)in rabbits.Results The expression of CD68,CD43 and actin in aortic and coronary plaques was no remarkably different ( P >0.05).The positive materials of CD68 were mostly diffuse distribution and the positive materials of CD43 were primarily central distribution in fatty streak plaques.The expression of CD68,CD43 in fatty streak plaques were remarkably higher than that in fibrous plaques ( P <0.01).Actin in fibrous plaques were significantly higher than that in fatty streak plaques ( P <0.01).Conclusions It is suggested that the foam cells of atherosclerosis originated mostly from macrophages in the streak plaques, and the origin of foam cell is mostly VSMC in fibrous plaques and it is related to formation of fibrous cap in atherosclerosis plaques.The interaction of T lymphocytes and macrophages,in locally infiltration may be a major element in fatty necrosis of plaques.
