部分栽培葱属植物叶绿体基因组的RAPD分析

【摘要】 提取我国栽培的几种主要葱属植物的叶绿体DNA ,利用RAPD技术进行遗传关系分析。从 5 6个 10bp随机引物中筛选出 18个多态性引物进行扩增分析 ,14个材料共扩增出2 70条带 ,其中 2 4 6条具多态性。UPGMA聚类将供试葱属品种分成 4个组。葱属在进化上存在两个方向 ,分别形成了管状叶和扁平叶两种类型

【Abstract】 Fourteen Allium cultivars were analyzed by RAPD technique to determine their genetic relations. Of the 56 decamer primers screened, 18 showed consistent band pattern and amplification, and produced 270 bands with 246 polymorphic bands in the chloroplast genomes. The genetic distances among the materials ranged from 0 to 13.2. Four sections were classified by UPGMA cluster analysis, which demonstrated that welsh onion chive and bunching onion should be divided into Fistulosum, bulb onion and shallot be clustered into Cepa, the others belong to Porrum and Rhiziridium. There might exist two directions in Allium evolution, pipy leaf type and flat leaf type of Allium were formed.
