核苷磷酸化酶的产酶条件优化的研究
Study on Optimal Conditions of NPase
【摘要】 以鸟苷和 5 氟尿嘧啶为底物 ,乙酰短杆菌菌体为酶源酶法合成 5 氟尿苷。采用酵母浸膏培养基(酵母浸膏 4.0 % ,葡萄糖 0 .4% ,NaCl 0 .3 % ,pH7.0 ) ,较高的溶氧量 ,同时添加 0 .6 %的豆油 ,培养 12h ,产酶量可提高 30 %。
【Abstract】 5-Fluorouride was enzymatically synthesized from guanosine and 5-fluorouracil by brevibacterium acetylicum The suitable culture medium contained 40g yeast extract, 4g glucose, 3g NaCl in 1 liter while pH was adjusted to 7 0 with NaOH After brevibacterium acetylicum was cultured for 12 h in the condition of the higher dissolve oxygen and 0 6% soybean oil supplied , the amount of enzym produced can be increased by 30%
