海葵等四种动物粘多糖碱提取的比较研究

【摘要】 采用碱提取法从绿疣海葵等 4种海洋无脊椎动物中提取酸性粘多糖 ,其粗品产率分别为 :绿疣海葵 9.9% ,绒毛近方蟹 4 .9% ,毛肤石鳖 4 .3% ,平背蜞 3.6% .用蒽酮比色法和酚 硫酸法测定产品糖含量 ,考马斯亮蓝法测定蛋白质含量 .醋酸纤维素薄膜电泳显示粘多糖的电泳迁移率相似 ,海葵粘多糖与甲苯胺蓝染色呈蓝色 ,绒毛近方蟹、平背蜞粘多糖呈紫色 .

【Abstract】 By means of alkaline extractive method, glycosaminoglycans are extracted from four marine invertebrates: sea anemone Anthopleuar midori (Uchida), chiton Acanthochiton rubrolineatus (Lischke) and two crab Gaetice depressus (De Haan), Hemigrapsus penicillatus (De Haan). The crude yield rates are: (9.9±0.1)%, (4.3±5.1)%, (3.6±0 7)%, (4.9±0.4)%, respectively. The reactions of saccharide with Anthron and phenol_sulphate acid are used to analyze saccharide content. The content of protein in the products is determined by Coomassie Brilliant Blue method. The results of cellulose acetate membrance electrophoresis (stained in 0.5 % toluidine blue) show the electrophoresis removal rates of these glycosaminoglycans are similar, the color of the glycosaminoglycan (stained in 0.5% toluidine blue) from A. midori (Uchida) is blue, but others are purple.
