用激光诱导荧光结合毛细管电泳测定单胺类神经递质
Determination of Amine Neurotransmitterswith Capillary Electrophoresis Coupled Laser-induced Fluorescence Detector
【摘要】 建立了用半导体激光诱导荧光结合毛细管电泳测定单胺类神经递质 ,去甲肾上腺素和多巴胺的高灵敏的分析方法。考察了青色素衍生物 (Cy5 )对去甲肾上腺素和多巴胺的各种衍生条件。在优化条件下 ,去甲肾上腺素和多巴胺在 3× 10 - 8~ 5× 10 - 6 mol/ L浓度范围内与其荧光强度呈现良好的线性关系。去甲肾上腺素和多巴胺的最低检测浓度分别为 5 .9× 10 - 9,5 .4× 10 - 9mol/ L。本方法的优点是简便 ,灵敏 ,样品用量少。适合于痕量单胺类神经递质的分析
【Abstract】 A sensitive,rapid and accurate analysis of the amine neurotransmitters was accomplished by capil- lary electrophoresis with a diode laser- induced fluorescence detector.Two important amine neurotrans- mitters,noradrenaline and dopamine were chosen as model compounds,and derivatized with Cy5 .The effects of some experimental conditions on the derivatization and separation were investigated in detail.The fluores- cence intensity of the derivative has a linear relationship with amine concentration between 3× 10 -8~ 5× 10 -6 mol/ L .The minimum concentration of their detection limits were 5 .9× 10 -9,5 .4× 10 -9mol/ L ,respectively.
