生物反应罐Vero细胞微载体灌流培养

【摘要】 用 7L生物反应罐 ,3g/LCytoedx 1,φ =5 0 %溶解氧 ,40~ 5 0r/min搅拌速度进行细胞培养 ,培养 48h开始灌流 .连续动态测定葡萄糖含量、渗透压变化及 pH值 ,同时观察细胞形态、增殖情况 .连续 3批 7L规模灌流培养表明 ,细胞贴附率高 ,增殖速度快 ,细胞形态良好 ,细胞群体倍增数达 4.7.结果表明生物反应罐灌流培养法是一种快速、规模化制备细胞的有效方法

【Abstract】 The cells was cultured in 7?L bioreactor with the microcarrier of 3 g/L,dissolved oxygen of 50% and stirring speed of 40—50?r/min.The perfusion was performed after 48?h culture.The dynamic glucose concentration,fluctuations of osmotic pressure and pH value was monitored continuously.Meanwhile,the cell and proliferation were observed.Three consecutive batches in 7?L perfusion culture demonstrated high,quick cell proliferation speed,good cell adhesions and appearances,and cellular population doubling level reached 4.7.The perfusion culture approach in bioreactor is a quick way to prepare cells in large scale.
