大鼠血浆TNF-α的测定以及小剂量rTNF-α对感染的大鼠保护作用
Determination of the level of TNF-α in plasma of rats and the protection of low dose of rTNF-α anginst infection
【摘要】 目的 测定烧伤早期大鼠和脓毒症感染大鼠血浆肿瘤坏死因子 α(TNF α)水平和丙二醛 (MDA)水平的变化 ,并探讨小剂量 (10 5U/kg)肿瘤坏死因子α(TNF α)对感染的保护作用。方法 用背部烫伤法和盲肠结扎穿刺法 (CLP)建立大鼠烧伤模型和内源性感染模型 ,用试剂盒测定血浆TNF α和MDA水平。结果 烧伤早期大鼠血中TNF α和MDA含量升高 (P <0 0 1) ,且升高程度与烧伤面积有关 ;脓毒症感染大鼠血中TNF α和MDA含量亦升高 (P <0 0 1) ;预先注射小剂量rTNF α可提高感染大鼠的存活率 (P <0 0 1)。结论 烧伤和脓毒症感染时血中TNF α水平明显升高 ,而预先注射小剂量rTNF α可使脓毒症感染发生率下降 ,动物存活率增加
【Abstract】 Objective To determine the levels of tumor necrosis factor-α(TNF-α) and malondialde-hyde (MDA) in plasma and to study the protection of low dose of rTNF-α anginst infection. Methods The rat models for sepsis and burn were established and ELISA was used to determine the levels of TNF-α and MDA. Results In burn model, TNF-α levels and MDA levels were correlated with the extent of the burn. Another study show that rats pretreated with low dose of rTNF-α had significantly higher survival rates (P<0.01). Conclusion Plasma TNF-α level increase in burn and sepsis. Pretreatment with a single low dose of rTNF-α prevents subsequent sepsis lethality.
