甘糖酯对大鼠尿激酶的诱导作用
The induction of urokinase(UK) in rat by propylene glycol mannite sulfate (PGMS)
【摘要】 目的 观察甘糖酯对大鼠尿激酶的诱导作用 ,从基因水平上探讨甘糖酯的抗栓机制。方法 采用发色底物法检测甘糖酯对大鼠血浆中尿激酶活性的影响 ,以半定量RT PCR法检测大鼠尿激酶基因表达。结果 大鼠经灌喂甘糖酯后血液中尿激酶活性和mRNA表达水平均高于对照组 ,且尿激酶活性和mRNA表达水平的升高同喂药剂量正相关 ,在 2 5~ 10 0mg·kg-1的剂量范围内随剂量增大而升高。结论 甘糖酯可以在mRNA水平上诱导大鼠尿激酶的表达 ,或加快大鼠尿激酶mRNA的转录速度或增加mRNA的稳定性 ,激活纤溶系统 ,从而表现出一定的抗栓活性
【Abstract】 AIM to observe the induction on UK by propylene glycol mannite sulfate (PGMS) in rat blood and try to reveal the mechanism of antithrombotic of PGMS on gene level. METHODS The activity of UK was measured by chromogenic method before and after giving PGMS to rat and the expression of UK mRNA were quantified by semi quantitative RT PCR. RESULTS the UK activity of experimental group and the expression of UK mRNA are higher than that of control group. The increase of UK activity and mRNA expression is in a dose dependent manner after giving PGMS to rat within the dose of 100mg·kg -1 . CONCLUSION PGMS can induce UK gene expression. This increase of UK gene expression may therefore reflect, in part, an increase in the stability of UK mRNA and/or processing of nuclear UK transcription, which in turn lead to the increase of UK activity in rat blood. As result,PGMS can activate the fibrinolytic system and have function of antithrombotic.
