硫代硫酸钠对顺铂所致肾毒性中和作用的实验研究

【摘要】 目的 进一步探讨硫代硫酸钠 (STS)对顺铂 (DDP)所致肾毒性的中和作用的实验研究。方法 小鼠随机分为阴性对照组 (生理盐水组 )、阳性对照组 (DDP组 )、DDP +STS组 ,各组均每隔 2d给药一次 ,3 3d后观察肾功能 ,即尿素氮(BUN)和肌酐 (Cr)的变化 ,超氧化物歧化酶 (SOD)和谷胱甘肽过氧化物酶 (GSH -Px)活性改变 ,肾脏病理。结果 DDP组血清中BUN升高与生理盐水组比较差异有显著性 ;加入STS以后血清中BUN的值与DDP组比较差异有显著性。Cr结果显示 :DDP组与Cr值比较差异有显著性。SOD和GSH -Px结果显示 :DDP组、DDP +STS组与生理盐水组比较差异有显著性。肾脏病理组织学结果表明 :STS比较明显减轻了DDP所致的肾毒性 ,其中BUN、Cr和SOD、GSH -Px发生改变。结论 STS能在一定的程度上缓解顺铂所致的肾毒性

【Abstract】 Objective To study the effect of sodium thiosulfate (STS) on renal’s function (urea nitrogen and creatinine), the activities of superoxide dismutase (SOD) and glutathione perxidase (GSH-Px) and change of renal’s histopathology in experimental mice.Methods Mice were divided randomly into three groups, namely the control group, DDP (cisplatin) group and the DDP+STS one. The change of renal’s function as well as histopathology were observed, the change of activities of SOD,GSH-Px were also detected. Results The content of serum BUN and Cr of DDP group were significantly higher than that of control group and significant decrease was shown in DDP+STS group compared with DDP group. The activities of SOD,GSH-Px were lower in DDP+STS group than those of DDP group. The main damage caused by cisplatin could be observed as the severe tumefaction of epithelium cells of convoluted renal tubules,but rather medium tumefaction in DDP+STS group.Conclusion STS could alleviate the renal’s damage caused by cisplatin in some degree.
