高温与电离辐射联合对鼠胎脑热休克蛋白及蛋白质的影响

【摘要】 目的 探讨高温与电离辐射联合对鼠胎脑热休克蛋白 (HSP70 )和蛋白质的影响。方法 孕d1 0的小鼠给予 4 2℃和 1 .0Gy( 6 0 Co)处理 ,取胎鼠全脑 ,用Westerndotblot和Lowrey方法测量HSP70和蛋白质含量。结果  1 .0Gy照射组、照射联合高温 ( 42℃ )组与对照组比较胎脑蛋白质明显减少 ,并诱导HSP70合成。高温 ( 42℃ )联合照射组与 4 2℃组相比胎脑蛋白质含量没有显著性变化 ,但二组诱导HSP70合成能力强。结论 高温诱导合成HSP70能力较辐射强 ,辐射引起胎脑蛋白质含量减少 ,预先高温处理孕鼠 ,然后再给电离辐射 ,胎鼠表现出交叉适应性反应

【Abstract】 Objective To study if hyperthermia and ionizing radiation could produce combined effects on heat stress protein(HSP)and protein of embryonic brains of mice. Methods The mice with gestation of 10 day were exposed to temperature of 42℃ and 1.0Gy 60 Coγ-rays.The content of HSP70 and protein of embryonic brains of mice was measured with Western dot blot and Lowrey method.Results The contents of protein of embryonic brains in the 1.0Gy group and in combined 1.0Gy with 42℃ temperature group were lower than those of the control,and these two groups may induce the synthesis of HSP70.However,there’s no siginficant difference in contents of protein of embryonic brains of mice between combined 42℃ temperature with 1.0Gy group and 42℃temperature group,but the contents of HSP70 increased signficantly in embrymic brain of mice in these two groups.Conclusion The effect induced by hyperthermia on synthesis of HSP70 is higher than that of the ionizing radiation.The ionizing radiation could reduce the contents of protein of embryonic brains of mice.Pre-exposure to the temperatuer of 42℃ for 5 mins followed by 1.0Gy 60 Co γ-ray radiation could induce cross-adaptive response of embryonic brain of mice.
