变色酸2B分光光度法测定蛋白质

【摘要】 在pH值为 1 2~ 2 0的Clark Lubs缓冲溶液中 ,变色酸 2B与蛋白质结合形成复合物 ,导致变色酸 2B褪色 ,其最大褪色波长位于 5 3 0nm .不同蛋白质在 0~ 3 0 μg/mL至 0~ 5 0 μg/mL范围内服从Beer定律 ,其表观摩尔吸光系数视蛋白质的不同分别在 1 1× 10 5~ 1 2× 10 6 L·mol-1·cm-1之间 .方法不仅具有高灵敏度 ,而且选择性好、简便快速 .用于人血清样品中总蛋白质量的测定 ,结果满意 .

【Abstract】 A highly sensitive fading reaction has been developed, based on the fact that Chromotrope 2B reacts with proteins to form complexes in Clark-Lubs buffer with pH 1.2-2.0 and fading of Chromotrope 2B takes place. The maximum fading wavelength was at 530 nm. Beers law was obeyed in the range from 0-30 μg/mL to 0-50 μg/mL depending on the different proteins. The reactions had highly sensitivities and their apparent molar absorptivities of fading reactions were between 1.1×10 5 L·mol -1 ·cm -1 and 1.2×10 6 L·mol -1 ·cm -1 depending on the different proteins. The method was highly selective, rapid and simple and has been applied for spectrophotometric determination of total amounts of proteins in human serum samples with satisfactory results.
