藻菌类毒性分解代谢物的分离与鉴定
ISOLATION AND IDENTIFICATION OF TOXIC METABOLITE FROM PHYCOMYCETES
【摘要】 :本文报道微小根毛霉 (Rhizomucorpusillus) ,足样根霉 (Rhizopusrhizodiform)和伞枝犁头霉(Aosidiacorumoifera)接种于已灭菌的草木犀粉 ,胡麻饼和玉米粉培养基中。于 37℃培养 15d后 ,提取培养物所得分馏物 ,经分离与鉴定 ,获得 3种毒性产物。 (1) 3种菌在草木犀培养物中分离到香豆素和双香豆素 ,而胡麻饼和玉米粉培养物中未分离到上述产物。这表明香豆素和双香豆素并非藻菌的代谢产物 ,而是由于藻菌腐生于草木犀后的分解产物 ,使其草木犀自身含有的香豆素分解转化成双香豆素 ,并使其含量成倍增长 ,导致牛、羊中毒。 (2 )以标准黄曲霉毒素B1通过薄层层析的确证试验 ,确认足样根霉在玉米粉培养基中能产生黄曲霉毒素B1。 (3) 3种菌在 3种培养基中同时分离到 16、18碳直链固态饱和脂肪酸。 16碳脂肪酸分子式CH3 (CH2 ) 14 COOH ,分子量2 5 6 1,18碳脂肪酸分子式CH3 (CH2 ) 16 COOH ,分子量 2 83 4。 16、18碳脂肪酸是藻菌在上述培养基中形成的分解产物 ,致使基质中脂肪分解 ,含量降低 ,而脂肪酸含量大增 ,它反应了饲料霉变的本质 ,也是动物酸中毒的成因。
【Abstract】 Rhizomucor pusillus,Rhizopus rhizodiform and Absidia corumoifera were inoculated in clover,flax cake and maize powder and cultured at 37 ℃ for 15 days.Three kinds of products were obtained by extracting,isolating and identifing the culture media.(1)Coumarin and dicoumarin were isolated from the three fungi inoculated clover culture medium,but were not isolated from flax cake and maize powders.The results show that coumarin and dicoumarin are not metabolite of phycomycetes,they are decomposition products of clover polluted by phycomycetes.Coumarin of clover is decomposed to dicoumarin,resulting in double increases of its content and intoxication of cattle and goats.(2)It was confirmed that aflatoxin B 1 could be produced in Rhizopus rhizodiform cultured maize powder with thin layer chromatography using standard aflatoxin B 1.(3)16,18 C straight chain solid state staturated fatty acids were isolated from all the three kinds of culture media inoculated the three kinds of fungi.The molecular formula of 16 C fatty acids was CH 3(CH 2) 14 COOH,its molecular weight was 256.1.The molecular formula and molecular weight of 18 C fatty acids were CH 3(CH 2) 16 COOH and 283 4,respectively.16 and 18 C fatty acids were decomposition products from the above three kinds of culture media inoculated Phycomycetes,leading to fat decomposition and its content decrease in the media.But the contents of fatty acids increased greatly.These reflect the nature of feed moulding and are the cause of acid intoxication in cattle and goats.
