亚胺环己酮对猪卵母细胞人工孤雌激活作用的研究
ARTIFICIAL ACTIVATION OF PORCINE OOCYTES MATURED IN VITRO
【摘要】 本文以体外成熟的猪卵母细胞为材料 ,以乙醇、电脉冲和氯化锶为人工刺激条件 ,研究了蛋白质合成抑制剂———亚胺环己酮 (CHX)对猪卵母细胞的孤雌激活效果的影响。结果表明 ,乙醇、电脉冲和SrCl2 均可使猪卵母细胞激活 ,而电脉冲的激活效果最好。当分别与CHX联合使用时 ,激活率显著高于单独的乙醇、电脉冲或SrCl2 刺激。说明 ,CHX与乙醇、电脉冲及SrCl2 联合使用对猪IVM卵母细胞激活具有显著的协同促进作用。
【Abstract】 This study was designed to evaluate the effects of cycloheximide (CHX),a protein synthesis inhibitor,on artificial activation of in vitro matured (IVM) porcine oocytes with alcohol,electric pulse or strontium cholide (SrCl 2),respectively.The results showed that alcohol,electric pulse or SrCl 2 could activate the porcine oocytes which were cultured in modified TCM 199 media for 44 48 h.When the oocytes were primed with alcohol,electric pulse or SrCl 2 and then cultured in CHX contained TCM 199 media for 20 h,the activation rates were significantly higher than that stimulated with alcohol,electric pulse or SrCl 2 alone.In conclusion,alcohol,electric pulse and strontium chloride acted synergistically with CHX in activating the porcine IVM oocytes.
