反相高效液相色谱法测定血浆中两性霉素B浓度

【摘要】 目的 :采用反相高效液相色谱法测定两性霉素B(AMB)的血药浓度 ,用于该药的治疗药物监测。方法 :在ODS -HypersilC18柱上 ,以 0 .0 0 5mol/LEDTA - 2Na溶液 -甲醇 (2 :8v/v)为流动相 ,流速为 1.5ml/min ,检测波长为 380nm ,测定血中AMB浓度。结果 :AMB保留时间为 3.85min ,在 0 .12 5 - 4.0 μg/ml之间有较好的线性范围 ,r=0 .9998,最低检测浓度为 0 .0 6 μg/ml,AMB样品日内精密度和日间精密度均小于 3 .5 % ,平均方法回收率 99.4%。结论 :本方法专一性强 ,回收率高 ,样品保留时间短 ,适用于临床AMB的治疗药物浓度监测。

【Abstract】 OBJECTIVE:To establish a reversed-phase HPLC assay to monitor the plasma concentration of Amphotericin B.METHOD:Amphotericin B was separated on ODS-Hypersil C 18 column and detected at UV380nm,using 0.005mol/L EDTA-2Na-methanol(2:8,v/v) as mobile phase,and the flow rate was 1.5ml/min.RESULTS:The retention time of Amphotericin B was 3.87min and the standard curve was linear over the range of 0.125~4.0μg/ml,the correlation coefficient was 0.9998,the lowest detectable concentration was 0.06μg/ml, the coefficient variation of within and between-day were less than 3.5%,and the average recovery of the method was 99.4%.CONCLUSION:The method is senstive and specific with high recovery rate and short rerention time. It is suitable to monitor the plasma drug concentration in clinical Amphotericin B therapy
