新鲜自体骨移植界面愈合过程中钙、磷、硫元素含量的动态分析研究
Quantitative microanalysis of Ca,P and S elements concentration on the interface calcification process after fresh autogenous bone transplantation
【摘要】 目的 通过在日本大耳白兔下颌骨体形成的人工缺损中进行新鲜自体骨移植 ,研究移植骨———宿主骨皮质界面骨性愈合过程中钙、磷、硫三种元素的浓度变化 ,以揭示移植界面愈合的基本规律。方法 利用X射线能量衍射微区分析法、改进的断面冷冻抛光和峰背比计量技术 ,进行移植界面三种元素 2 4周动态定量检测。结果移植界面钙、磷含量在术后 2~ 3周时明显低于正常对照 (P <0 .0 1) ,但随后逐渐增加 ;而同期硫含量则显著高于正常水平 ,其后迅速下降 ;在 5周时 ,界面骨质中的钙、磷、硫元素含量已接近正常状态 (P >0 .0 5 ) ,但各微区数值差异较大 ;8~ 12周后 ,界面骨质钙、磷元素均匀分布 ,元素含量变化趋于稳定。结论 骨移植初期 ,界面骨质有明显的吸收过程 ;在移植 5周时 ,移植骨与宿主骨界面就可形成较高钙化程度的骨性连接 ,但矿化不尽完善 ;随后通过不断的应力改建 ,在移植 8~ 12周后 ,可望形成矿化程度更高的稳定界面结合。钙、磷元素含量在移植界面愈合中的变化高度相关 ,其动态变化基本可反映出骨移植界面愈合的基本进程
【Abstract】 Objective The present study was conducted for the interface calcification processes by detecting three elements’ concentration changes of Ca P S after fresh autogenous bones graft repairing the experimental defects on Japanese rabbits’ mandibular body.Methods These elements concestrations were observed continuously from 2 to 24 weeks by X-ray energy diffractive system (EDS),microanalysis of thin sections of frozen polishing and peak-background comparison methods.Results There were rapidly concentration decreases of Ca and P and increase of S during 3 weeks post-operatively,especially at 2 weeks (P< 0.02 ).Following these,the concentrations of Ca,P and S elements were gradually regained their pre-operative levels and nearly comparable to the conccntrations of normal cortical bone at 5 weeks (P> 0.05 ).But the distributions of interface inorganic salts were not equall at the same time.However,the differences of Ca and P concentrations between normal cortical bones and autografted interfaces would be faded away after 8 or 12 weeks.Conclusion There is obviously an absorption process of the interface bones in the primary stage after transplantation.But the dense union between host and graft bones forms at 5 weeks and the remodeling generally is achieved at 8 or 12 weeks.This study indicates that the concentration changes of Ca and P correlate with the processes of interface mineralization.
