一种新的鉴定水稻初级三体的方法(英文)

【摘要】 水稻初级三体在水稻遗传育种中起着重要的作用。其归属鉴定通常用 Shastry(1960 )的粗线期核型分析方法 ,但这种方法在实际操作中有一定的难度 ,不易得到分散好的粗线期染色体分裂相。本研究首次尝试利用三体株和正常株同源染色体间 DNA含量的差异 ,用分子标记的手段将量的差异转为自显影带的差异原理去鉴定三体 ,发现此鉴定结果与粗线期核型分析结果一致 ,而此方法在实验技术上可靠性高 ,快捷 ,因此是一种新的有效鉴定初级三体的方法

【Abstract】 The primary trisomics of rice play an important role in study of genet ics and breeding of rice. Researchers always identify primary trisomics by the k aryotype analysis of pachtene firstly used by Shastry(1960). But this method is not easy to obtain good result because it needs superb skill. In this study, a n ew way to identify primary trisomics was used. It makes use of the difference in the quantity of homologous DNA between primary trisomics and diploid rice and d emonstrated the difference with molecular marker. The identification result by m olecular marker is the same as that by karyotype analysis. This method is credib le and quick, so it will be a new and useful means to identify primary trisomics
