碳及氮源在提高重组酵母表达分泌水平中的作用
The effects of carbon sources and nitrogen sources on raising the expression and secretion of α-amylase in recombinant yeast
【摘要】 通过摇瓶批式发酵方法和 2 L发酵罐补料分批发酵方法研究了碳及氮源在提高重组酵母 S.cerevisiae2 0 B- 1 2 ( p NA3)表达分泌水平中的作用 ,得出采用分段发酵方法 ,在诱导期补充替代碳源(乳糖或甘油 )并以营养丰富的天然氮源 (酵母膏 )诱导产物分泌可使重组酵母表达、分泌水平有较大提高。在补料分批培养中通过生长阶段加速补充碳源 ,产酶阶段补充替代碳源和分次补加酵母膏的方法使重组酵母表达、分泌水平进一步提高 ,从而为流加培养和工业化放大提供了依据。
【Abstract】 The effects of carbon sources and nitrogen sources on raising the expression and secretion of α amylase incombinant yeast was studied. The results show that: by using two stage cultivation system, feeding right carbon source timely in the growth stage, feeding the alternate carbon source(lactin or glycerin) and using the natural nitrogen to induce the secretion of the production in the production stage, the level of the expression and section in recombinant yeast reaches higher level. In the fed batch fermentation, by accelerated feeding the carbon source in the growth stage, feeding the natural nitrogen source and alternate carbon source timely in the production stage ,the expression and secretion of α amylase reach much higher levels.
