Vc二步发酵中伴生菌的作用机制
THE EFFECT OF BACILLUS MEGATERIUM IN VITAMIN C TWO-STEP FERMENTATION
【摘要】 应用细胞培养和膜分离技术研究了Vc两步发酵中伴生菌巨大芽孢杆菌 (Bacillusmegaterium)对氧化葡萄糖酸杆菌 (Gluconobacteroxydans)产酸作用机制。结果表明 :巨大芽孢杆菌培养液中分子量在 30~ 5 0kD及大于 1 0 0kD组分明显促进产酸 :其组分通过凝胶层析分离纯化 ,自动紫外检测仪检测 ( 2 80nm) ,聚丙烯酰胺凝胶电泳及考马斯亮兰G2 5 0特异染色 ,初步证实为蛋白质 ,且至少是两种以上蛋白质 ,它们在低温下稳定性较好
【Abstract】 The effect of Bacillus megaterium (B.m) on 2-keto-L-gulonic acid (2-KGA) synthesis by Gluconobacter oxydans(G.o) was studied in Vc two-step fermentation process.By using ultrafiltration,Bacillus megaterium culture the supernatant from B.m was divided into several parts in molecular weight.Of which the metabolites with molecular weight range from 30,000 to 50,000 Dalton and that over 100,000 Dalton can enhance the ability of G.o in converting L-sorbose into 2-KGA apparently.Further isolating and purifying the 30,000~50,000 Dalton and over 100,000 Dalton by means of Gel Chromatography,detection by auto-ultraviolet detector,polyacrylamide gel electrophoresis and specific dying by Coomassie brilliant blue G250,the materials could be at least two sorts of proteins which are stable at low temperature (4℃).
