6-巯基嘌呤与2-巯基嘌呤在银电极上的伏安行为的比较
Comparison of Voltammetric Behavior of 6-Mercaptopurine with that of 2-Mercaptopurine at a Silver Electrode
【摘要】 首次对 6-巯基嘌呤 ( 6MP)和 2 -巯基嘌呤 ( 2 MP)在银电极上的伏安行为进行了比较研究 ,并同时对它们的衍生物的电化学性质进行了探讨 .结果发现 ,6MP和 2 MP均可在银电极上产生氧化还原波 .当电位扫描范围较小时 ,不论是在酸性、中性还是碱性溶液中 ,它们各产生一对不可逆波 ,此为巯基银的形成和还原所致 .当电位扫描范围拓展至足够负时 ,6MP的伏安图上出现多个波 ,这是由于 6MP在较负电位处发生还原 ,其还原产物吸附在电极表面并进一步发生氧化还原所致 .6MP在此过程中失去巯基 ,随后被还原成 1,2 ,3 ,6-四氢嘌呤或 1,6-二氢嘌呤 .其电解产物的毛细管电泳分离结果表明 6MP确被进一步还原 .2 MP在相同条件下不产生多波现象 ,这与其不易脱巯基有关 .本文还对 6-巯基 - 2 -羟基嘌呤 ( 6TX)、2 -氨基 - 6-巯基嘌呤 ( 6TG)、2 -巯基 - 6-羟基嘌呤 ( 2 TX)和 6-巯基嘌呤核苷 ( 6MPR)等进行了研究比较 .
【Abstract】 6-mercaptopurine (6MP) as well as 2-mercaptopurine (2MP) can generate an irreversible wave at a silver electrode in sodium phosphate buffers with different pH value. The wave results from the formation and reduction of silver thiopurine thiolate and can be used for the cathodic stripping voltammetry of them. When the potential scan-range extends to sufficient negative, 6MP exhibits multiwave due to the reduction of 6MP and its reduction products adsorbed further. During the electrochemical process, 6MP losses -SH and is reduced to 1,2,3,6-tetrahydropurine. Capillary electropherograms of 6MP and electrolyzed 6MP support this conclusion to some extent. 2MP does not exhibit similar phenomenon to 6MP under the same conditions. In this work, 2-thioxanthine, 6-thioxanthine, 2-amino-6-mercaptopurine and 6-mercaptopurine riboside are also tested and discussed.
