【摘要】 目的　研究“益肝浓缩煎剂”、丹参复方 (丹参 +当归 +水蛭 )、当归、水蛭对大鼠肝纤维化预防与治疗作用 ,并对其机制进行初步探讨。方法　以CCl4诱导大鼠肝纤维化模型 ,造模前 2周及造模结束后分别予相应药物进行防治 ,各持续 10周 ;测定肝组织羟脯氨酸 (Hyp)、丙二醛 (MDA)含量 ,光镜、电镜观察肝脏病理形态变化。结果　“益肝浓缩煎剂”预防、治疗组Hyp、MDA显著降低 (P <0 .0 5 ) ,胶原沉积及脂肪变性明显减轻 (P <0 .0 5 ) ;丹参复方治疗组Hyp、MDA及纤维增生均显著下降 (P <0 .0 5 ) ;当归治疗组Hyp、MDA显著下降 (P <0 .0 5 ) ,胶原沉积无减轻 (P >0 .0 5 ) ;水蛭治疗组各项指标均无好转 (P >0 .0 5 ) ;“益肝浓缩煎剂”预防组α 平滑肌肌动蛋白染色强度减弱。结论　“益肝浓缩煎剂”对大鼠肝纤维化具有明确防治作用 ;丹参复方对大鼠肝纤维化具有明确治疗作用 ,当归次之 ,仅Hyp、MDA显著下降 ,水蛭作用较差 ;上述疗效作用机制可能与抗脂质过氧化或抑制肝星状细胞活化有关。更多还原

【Abstract】 Aim To study the preventive and therapeutic effects of Concentrated Yigan Decoction,formula of Radix Salviae Miltiorrhizae(i.e. Radix Salviae Militiorrhizae+Radix angelicae Sinensis+Hirude),Radix Angelicae Sinensis?Hirude on hepatic fibrosis in rats induced by carbon tetrachloride(CCl 4) as well as their relative mechanisms.Method Hepatic fibrosis in rats was induced by CCl 4 and the relative herbs were given 2 weeks before (prevention group) or after model production(treatment group)respectively.The administration of the herbs lasted for 10 weeks.Liver hydroxyproline(Hyp) and malondialdehyde(MDA)level were determined,and histopathological changes were observed by optical microscopy and electromicroscopy.Results Hyp,MDA,hepatic fibrosis,and degrees were markedly resduced respectively compared with those of control group in Concentrated Yigan Decoction prevention and treatment group( P <0.05).The contents of Hyp and MDA as well as degrees of liver fibrosis in formula of Radix Salviae Miltiorrhizae treatment group were markedly lower than those of model of natural recovering group ( P <0.05).Hyp and MDA were markedly ( P <0.05)reduced in Radix Angelicae Sinensis treatment group while degrees of liver fibrosis had no statistical change ( P >0.05).In Hirude group there was no significant decrease in all respects ( P >0.05).The expression of α smooth muscel actin was reduced in Concentrated Yigan Decoction prevention group.Conclusions (1)Concentrated Yigan Decoction had remarkably preventive and therapeutic effects on hepatic fibrosis of rats.(2)Formula of Radix Salviae Miltiorrhizae could resolve ECM components accumulated in liver of rats,superior to Radix Angelicae Sinensis.Hirude had no therapeuti effect.(3)Antiperoxidation of lipid and depression of HSC activation might be related to the herbs preventive and therapeutic effects.更多还原
