海水养殖鱼类两种新贝尼登虫的RAPD分析
Random amplified polymorphic DNA (RAPD) analysis of two Neobenedenia species from cultured marine fishes
【摘要】 本文采用RAPD技术对寄生在福建沿海养殖大黄鱼和高体鱼师鱼体上的鱼己新贝尼登虫和梅氏新贝尼登虫的DNA多态性进行分析 ,结果显示 :①两者具有高度的遗传相似性 ( 83.1 %~ 94.7% ) ,形态观察和分子检测结果相印证 ,两者有可能为同种异名 ;②不同海区鱼体表的寄生虫表现出一定的差异和分化 ,但相同宿主虫体的相似性都在 90 %以上 ;③虫体睾丸边缘光滑与否等形态特征应为种内变异 ,不宜作为分类标准
【Abstract】 New evidence through RAPD analysis was provided for the controversial classification of Neobenedenia (Monogenea: Capsalidae) in this study. 15 arbitrary 10 nucleotide primers were used to detect the polymorphism of genomic DNA of Neobenedenia girellae and N. melleni parasitized on Seriola dumerili and Pseudosciaena crocea farming in net cages in Xiamen, Lianjiang and Putian of Fujian Province. The great genetic similarity index (83.1%~94.7%) indicated that there is little obvious genetic differentiation between N. girellae and N. melleni. The scarcity of clear morphological identification and the small genetic distance (5.3%~16.9%) indicate that N. girellae , which was formerly described as a separate species, may be synonym of N. melleni .
