维拉帕米抑制原代培养兔视网膜色素上皮细胞增殖的实验研究

【摘要】 为评价维拉帕米 (verapam il)对兔视网膜色素上皮细胞 (retinal pigment epithelium,RPE)增殖的影响 ,在原代培养的兔 RPE中分别加入不同浓度的维拉帕米溶液 (0 .1μm ol/ L,1μm ol/ L,1× 10 1 μm ol/ L,1× 10 2 μm ol/ L,1×10 3 μmol/ L ,1× 10 4μmol/ L ) ,于培养 72 h后用四甲基偶氮唑盐 (MTT)比色法 ,在酶联免疫测定仪上测 6 30 nm处吸光值 ,同对照组相比 ,计算出不同药物浓度条件下的抑制率。结果表明维拉帕米在 0 .1μm ol/ L浓度以上对 RPE的增殖有抑制作用 ;在 10μmol/ L浓度时使 RPE增殖率显著下降。提示维拉帕米作为钙通道拮抗剂对 RPE的增殖有抑制作用 ,可作为今后临床上防治增殖性玻璃体视网膜病变 (PVR)的药物进行研究

【Abstract】 The effects of calcium channel antagonist,verapamil on in vitro proliferation of retinal pigment epithelial cells (RPECs) in rabbits were studied. RPECs of rabbit were in Dulbecco’s modified eagle medium (DMEM) supplemented with 10 % fetal bovine serum and different concentrations of verapamil (0 1, 1,10, 100, 10 3 ,10 4 μmol/L). After culturing for 72 h, the absorption values of the solution was determined by MTT method at 630 nm and compared with those in the control group. The proliferation rate at different concentrations of verapamil was calculated. The results showed that over 0 1 μmol/L of verapamil could inhibit the proliferation of RPECs. 10 μmol/L of varapamil could significantly decrease the proliferation rate. These data suggest that verapamil can inhibit the proliferation of RPECs and might be used as a medicine for preventing and treating vitreoretionopathy in clinical practice.
