小鼠肺和气道树突状细胞分布特点及地塞米松对其的影响
The Distribution of Dendritic Cell in the Lung and Conducting Airway of Mouse and the Change of Airway Intraepithelial Dendritic Cell After Exposure to Dexamethasone
【摘要】 目的 探讨小鼠肺和气道树突状细胞 (DC)分布特点及地塞米松对其的影响。方法 实验动物分为地塞米松组和对照组 2组。采用免疫组化染色技术检测肺部DC及计算机图象分析技术进行定量测定和透射电镜分析DC形态。结果 对照组小鼠的整个肺组织包括气管、支气管、肺泡和脏层胸膜构成一个完整的NLDC - 145 + DC网络 ,大气道到小气管的DC密度为 5 0 0± 70个 /mm2 (P <0 .0 5 )~ 6 0± 2 0个 /mm2 (P <0 .0 5 )气道内皮表面积 ,电镜下DC呈典型树突状形态 ;地塞米松组DC分布特点与对照组相似 ,但大气道到小气道的DC密度为 2 0 0± 30个 /mm2 ~ 40± 10个 /mm2 气道内皮表面积 ,与对照组相比 ,NLDC - 145 + DC数量明显下降 (P <0 .0 5 ) ,DC树突状形态不典型。结论 树突状细胞在整个肺组织构成一个完整的NLDC - 145 + DC网络 ,地塞米松下调肺组织DC数量 ,但不影响其在肺部的分布。
【Abstract】 Objective To study the distribution of dendritic cells in the lung and conducting airways of mouse and the change of airway intraepithelial dendritic cells after exposure to dexamethasone. Methods BALB/c mice were divided into two groups:dexamethasone-treated group and control group.Pulmonary and tracheal samples were collected.Rat anti mouse dendritic cell (NLDC-145),goat anti rat IgG-HRP and DAB-staining technique were used to detect DC,furthermore,transmission electron microscope and computer image analysis were applied in this study.Results Dendritic cells in tracheal bronchiole and alveolar whole mounts were revealed by horseradish peroxidase staining using the NLDC-145 monoclonal antibody.In control mice,a dense network of NLDC-145 cells with typical branch tree-like morphosis was observed (the large airway conducting to the small airway conducting:500±70cells/mm 2~60±20/mm 2 epithelial mucosa,mean±SEM,n=7).In mice treated with dexamethasone the dense network of NLDC-145 + DC cells was down-regulated (the large airway conducting to the small airway conducting:200±30cells/mm 2~40±10/mm 2 epithelial mucosa,mean±SEM,n=7).Compared to the control mice,the amounts of NLDC-145 + DC were obviously lowered and the morphosis of dendritic cells was non-typical.Conclusion Dendritic cells consist of a complete NLDC-145 + DC network in the whole pulmonary;dexamethasone down-regulates the amount of the dendritic cells but not change the dendritic morphosis of dendritic cells in murine lung.
