阿霉素对兔心功能及心肌一氧化氮合酶表达的影响

【摘要】 目的 探讨阿霉素对兔心功能及心肌一氧化氮合酶 (NOS)表达的影响。方法 测定实验组和对照组兔心输出量(CO)、左心室收缩压 (LVSP)、左心室舒张末压 (LVEDP)。取心肌组织进行HE染色和NADPH组织化学染色并进行图像定量分析 ,所测得数据均进行统计学处理。结果 实验组CO、LVSP均明显低于对照组 (P <0 .0 1) ,LVEDP明显高于对照组 (P <0 .0 1) ,心肌组织NADPH组织化学染色及图像定量分析结果显示 ,实验组NOS表达明显增强 (P <0 .0 1) ,且与CO呈负相关 (γ =-0 .83 0 4,P <0 .0 1) ,与LVSP呈负相关 (γ =0 .7815 ,P <0 .0 5 ) ,与LVEDP呈正相关 (γ =0 .73 5 6,P <0 .0 5 )。结论 NOS过度表达 ,导致NO生成过量 ,参与阿霉素慢性心功能损伤的过程。

【Abstract】 Objective To investigate the effect of adriamycin on heart function and expression of nitric oxide synthase.Methods The CO,LVSP,LVEDP of the experiment group and the control group were weasured.Histology,reduced form of nicotinamide-adenine dinucleotide phosphate(NADPH) histochemistry of myocardium and image analysis were performed.Data were made a statistical analysis.Results Compared with the control group,CO,LVSP were significantly decreased(P<0.01),but LVEDP was significantly increased(P<0.01) in the experiment group.The expression of NOS in the experiment group was significantly increased,and was negative related to CO,LVSP,positive realted to LVEDP.Conclusion The higher expression of NOS may increase the production of NO,and it plays a role during the progress of ADM-induced impairment of heart.
