脲酶结合邻甲酚酞络合剂显色检测血清及尿液尿素含量

【摘要】 目的 建立一种实用的血清和尿液中尿素含量的比色测定方法。方法 脲酶水解尿素产生氨(NH3) ,使溶液 p H升高 ,以邻甲酚酞络合剂 (OCPC)作为 p H指示剂 ,波长 570 nm吸光度变化与尿素浓度成正相关。结果 该法线性范围血清 1 .0 0~ 2 0 0 mmol/ L,尿液 1 0 .0~ 1 0 0 0 mmol/ L ;检测界限血清0 .36mmol/ L,尿液 2 .1 9mmol/ L。精密度分析 :血清批内 CV1 .1 3%~ 2 .78% ,批间 CV1 .81 %~ 3.91 % ,总 CV2 .1 3%~ 4.80 %。尿液批内 CV0 .68%~ 1 .72 % ,批间 CV0 .96%~ 2 .2 4 % ,总 CV1 .1 7%~ 2 .82 %。平均回收率血清 1 0 0 % ,尿液 1 0 2 %。该法 (Y)与酶速率法 (X)比较相关性良好 (血清 Y=1 .0 4 8X+0 .51 1 ,r=0 .998,SXY=0 .62 0 mmol/ L:尿液 Y=1 .0 4 4 X- 6.41 4,r=0 .997,SXY=9.368mmol/ L)。结论 该法灵敏、简便、准确、线性范围广 ,适合自动分析。

【Abstract】 Objective To create a simple and useful enzymatic method for determination of urea in serum and urine.Methods pH increase resulting from the ammonia released by urease hydrolysis of urea, O cresolphthalein complexone is used to monitor the pH change colorimetrically.The absorbance veriation at 570nm are linerly related to urea concentrations.Results The linearity was 1 00~200 mmol/L for serum and 10 0~1000 mmol/L for urine.The detection limit was0 36 mmol/L for serum and 2 19 mmol/L for urine.Within run CV ,between run CV and total CV were 1 13%~2 78%,1 81%~3 91% and 2 13%~4 80% for serum and 0 68%~1 72%,0 96%~2 24% and 1 17%~2 82% for urine.The average recovery was 100% in serum and 102% in urine.A good correlation was shown as this method( Y )compared with kinetic method( X )(serum: Y =1 048 X +0 511, r =0 998, S XY =0 620 mmol/L.urine: Y =1 044 X -6 414, r =0 997, S XY =9 386 mmol/L).Conclusion This assay was simple,sensitive,precise,and widly linearity,suitable for automation.
