用国产重组不同优势抗原研制丙型肝炎病毒抗体确认试剂
HCV Antibody Confirmation Test Kit Using Recombinant Antigens from Different regions Of HCV Genome
【摘要】 采用基因工程技术 ,构建了人 IL- 1β融合表达的原核表达载体 p BVIL1 ,在大肠杆菌中表达了丙型肝炎病毒不同区优势抗原 ,研制出了丙型肝炎病毒抗体酶联免疫确认试剂。四种抗原混合包被对国家40份阳性参比血清检出率为 39/4 0。 40份阴性参比血清的检测均未出现假阳性。探讨了确认试剂生产的各种最佳条件。
【Abstract】 We highly expressed many immunodominant antigens from different regions of HCV genome using an unique fusion expression vector pBVIL1 constructed by us.Based on these recombinant antigens, we formulated a HCV confirmation EIA system.When coated with four combined antigens the detection of our National Reference Panel was 39/40 for the positive sera and 40/40 for the negative control panel.Optimization technology was disscussed.
