全反式维甲酸抑制肝癌细胞β-连环素的表达
Downregulation of Cytoplasmic β-Catenin in SMMC-7721 Human Hepatocellular Carcinoma Cells by All-trans-retinoic Acid
【摘要】 全反式维甲酸 (ATRA)是人肝癌细胞株SMMC 772 1的分化诱导剂 ,刺激细胞向正常方向分化 .为了研究ATRA诱导细胞分化的机制是否与 β 连环素 ( β catenin)有关 ,用终浓度为 1 0 μmol LATRA刺激SMMC 772 1 ,发现胞浆内β 连环素明显降低 .进一步实验证实 ,ATRA下调胞浆内β 连环素和诱导β 连环素的mRNA降低有关 .同时 ,ATRA处理基本不会对E 钙粘附素 β 连环素中的β 连环素的量产生影响 ,但能增强细胞凝聚力 .结果提示 ,ATRA对两种形式的 β 连环素的调节机制是不一样的 ,ATRA诱导细胞分化的机制和β 连环素有关 .
【Abstract】 All\| trans \|retinoic acid(ATRA),a differentiation inducer,stimulated SMMC\|7721 human hepatocellular carcinoma cells to differentiate to benign direction.β catenin played an important role in both cell\|cell adhesion and growth factor signal transduction.In order to find the role of β catenin in the process of ATRA\|induced differentiation,the changes of β catenin was examined in SMMC\|7721 treated with 10 μmol/L ATRA.The cytoplasmic β catenin was decreased.This change was the result of downregulation of β catenin mRNA.The ability of cell aggregation strength was increased after treatment.But the E\|cadherin\|associated populations of β catenin did not change.This suggested that ATRA up\|regulated the function of E\|cadherin/catenin complex,and its mechanism had no relationship with the expression of E\|cadherin\|associated β catenin.So the ATRA\|regulations of two intracellular pools of β catenin were different.Taken together,these data suggested that β catenin took part in the process of ATRA\|induced differentiation in SMMC\|7721.
