线刀损毁大鼠内侧前脑束建立部分损伤的帕金森病模型
CONSTRUCTING A PARTIALLY LESIONED PARKINSON DISEASE MODEL OF RAT BY TRANSECTING THE MEDIAL FOREBRAIN BUNDLE WITH A WIRE KNIFE
【摘要】 采用线刀切断大鼠内侧前脑束造成多巴胺能神经元的慢性退行性改变。在手术后不同的时间 ,分别采用辣根过氧化物酶逆行示踪、酪氨酸羟化酶免疫组化、行为学测试和高效液相色谱分析等方法对大鼠黑质神经元损伤状况、动物的异常旋转行为和纹状体内多巴胺的含量进行检测。结果表明 ,应用线刀切断大鼠内侧前脑束能够成功地建立部分损伤的大鼠帕金森病模型
【Abstract】 The medial forebrain bundle of rats was transected with a wire knife to set up a model of chronic degeneration of dopaminergic neurons. The abnormal rotation of rats, the extent of the substantia nigra neurons damage and the amount of dopamine in striatum were investigated by behavioral test, horseradish peroxidase retrograde tracing, tyrosine hydroxylase immunohistochemistry and high performance liquid chromatography at different time after operation. The results indicated that the partial lesioned model of Parkinson’s disease can be successfully established by transecting the medial forebrain bundle of rats with a wire knife.
