套式PCR检测恶性疟原虫Pfcrt基因76号编码多态性及RFLP分析

【摘要】 目的检测恶性疟原虫氯喹抗性转运基因 ( Pfcrt) 76编码多态性及其用该多态性显示恶性疟原虫对氯喹反应性进行评价。方法用套式 PCR/RFLP技术检测现场采集的滤纸血滴上恶性疟原虫 crt基因 76编码的赖氨酸野生型多态性及苏氨酸突变型多态性 ,并对恶性疟原虫的氯喹反应性进行体内法测定。结果 34份滤纸血样中 ,32份的恶性疟原虫 crt基因 76号编码为苏氨酸 (突变型 ) ,突变型比例为 94 .1 2 % ;34份血样中 2 1份做氯喹抗性体内观察 ,该 2 1个病例中 1 4份血样恶性疟原虫 crt基因 76号编码为突变型 ,体内法观察同样显示恶性疟原虫对氯喹有抗性 ,两种方法显示恶性疟原虫对氯喹有抗性的符合率为 66.67%。结论套式 PCR/RFLP检测 Pfcrt基因 76号编码多态性 ,其方法简便、快速 ,但其多态性对氯喹反应性的提示尚需进一步评价。

【Abstract】 Objective To evaluate the method of detection of polymorphism of P. falciparum chloroquine resistant transport gene 76 codon and prediction for P. falciparum chloroquine reaction. Methods Blood drop on filter paper collected from endemic areas were detected by nested PCR/RFLP and the chloroquine reaction observed in vivo . Results 32 out of 34 samples were defined to be the mutant type of Pfcrt gene 76 coden with a mutant rate of 94.12%. 21 out of the 32 samples collected from malaria patients were defined to be chloroquine resistant in vivo with a coincidence rate of 66.67%(14/21). Conclusions Nested PCR/RFLP is rapid, simple and convenient to detect the polymorphism of Pfcrt gene 76 codon, but it is necessary to evaluate further the prediction for chloroquine reaction according to the polymorphism.
